The present invention relates to an antibody that specifically binds to the receptor binding domain (RBD) of the SARS-CoV-2 spike protein and its use. More specifically, an antibody or antigen-binding fragment thereof that specifically binds to the receptor-binding domain of the SARS-CoV-2 spike protein, a composition for diagnosis of SARS-CoV-2 infection comprising the antibody or antigen-binding fragment thereof, SARS-CoV -2 Infection diagnostic kit, pharmaceutical composition for preventing or treating SARS-CoV-2 infection, method of providing information for diagnosis of SARS-CoV-2 infection using the antibody or antigen-binding fragment thereof, the antibody or antigen-binding fragment thereof It relates to a polynucleotide encoding a polynucleotide, a vector containing the polynucleotide, and a cell transformed with the vector. The antibody or antigen-binding fragment thereof of the present invention can effectively inhibit the binding between the SARS-CoV-2 spike protein and the ACE2 receptor of the host cell by binding to the receptor-binding domain of the SARS-CoV-2 spike protein. It is useful for the diagnosis, prevention, and treatment of CoV-2 infection, and has the advantage of being effective against such variants as it can also bind to RBD variants. Accordingly, by using the composition, kit, and information providing method of the present invention, infection of SARS-CoV-2 can be effectively diagnosed, prevented, or treated. In addition, the use of the polynucleotide, vector and cell of the present invention makes it possible to easily produce the antibody or antigen-binding fragment thereof of the present invention capable of exerting the above-described excellent effects.
