A test pack for the determination of alkaline phosphatase in human serum or plasma comprises bottled reagents comprising (A), a 3M aqueous solution of 2-amino-2-methyl-1-propanol containing about 1% w/v disodium monophenyl phosphate and about 0.6% w/v 4- aminoantipyrine, having a pH of about 10; (B), a buffered aqueous solution pH about 10 and containing about 0.32% w/v sodium carbonate and about 0.17% w/v sodium bicarbonate; (C), a colour developer solution having pH about 10. and containing about 0.32% w/v sodium carbonate, about 0.17% w/v sodium bicarbonate and about 2.4% w/v potassium ferricyanide, and (D) a standard solution containing 0.1% w/v phenol in 0.1 NHCI. In carrying out the test, sample is incubated with a mixture of solutions (A) and B, the resulting solution is incubated with reagent (C), and the product is measured at 500 mu against a standard prepared by incubating a mixture of solutions C and D.
