Hydrazine-stabilized bilirubin colorimetric assay - by coupling of free or conjugated bilirubin with diazonium salts

Novel method of estimating bilirubin in biological fluids in which bilirubin is coupled with a diazo-cpd. and the coupled prodt. is assayed colorimetrically in an alkaline medium, hydrazine being added as stabiliser to the reaction medium after addn. of the diazo-cpd. at a concn. of =400 times, and pref. 600 times that of the diazo cpd. The colour developed after coupling is stabilised and a rigid time limit in the assay of free or conjugated bilirubin is avoided, excess diazonium salt present after coupling of the conjugated bilirubin is inhibited, and no adverse colouration is introduced to confuse the assay.
