ERYTHROCYTE SENSITIZED WITH ANTIBODY OF POLYSACCHARIDE OF HEMOLYTIC STREPTOCOCCUS

PURPOSE:To obtain sensitized erythrocytes by which hemolytic streptococcus inflections disease can be diagnosed surely at early stage of infection by a simple procedure, by coupling as antibody consisting of polysaccharide of hemolytic streptococci to erythrocytes by means of a coupling agent. CONSTITUTION:An animal such as a rabbit and a goat is immunized with a hermolyic streptococci, and an antibody of cells of hemolytic streptococci thus obtained is purified soecifically, then an antibody of polysaccharide of hemolytic streptococci (PS-antibody) is separated. Next, the PS-antibody is brought into contact with a solution of an erythrocyte (preferably a fixed erythrocyte) treated with a coupling agent (e.g. tannin), and coupled it to produce the erythrocyte sensitized with the PS-antibody. As the sensitized erythrocyte mentioned above is agglutinated by a polysacchride antigen of a hemolytic streptococcus, the polysacchride antigen is detected by reverse passive hemagglutination, the infectious disease of hemolytic streptococci can be diagnosed easily at the early stage.
