A method for preparing hepatitis B virus surface antigen is presented. Thus, saccharomyces cerevisiae pAH 203, which was cultured in Burk Holder minimal medium contg. 20 g/ml histidine at 30C for 24hr, was suspended in 50mM phosphate buffer (pH=7.2) contg. 1.2M sorbitol, 14mM 2-mercaptoethanol, and 100 g/ml zymolyase-60,000 and shaked at 30C for 30min to give spheroplast followed by suspending it in a 50mM phosphate buffer contg. 0.1% Triton X-100 to give yeast lysate.
