摘要

PURPOSE:To prevent the deterioration in activity of protease, by hydrolyzing a protein at a pH within the range of a specific value or above and isoelectric point of the protein or below in a method for linking the protease to a carrier having positively charged functional groups and hydrolyzing the protein. CONSTITUTION:An immobilized protease obtained by covalently bonding a crosslinking agent to a carrier having positively charged functional groups and covalently bonding protease thereto, ionic bonding the protease to a carrier having positively charged functional groups and treating the resultant carrier with a crosslinking agent or covalently bonding a crosslinking agent to a carrier having positively charged functional groups and further treating the carrier with a crosslinking agent is used to hydrolyze a protein. In the process, the hydrolysis is carried out at a pH within the range of 3 or above and isoelectric point of the protein or below to prevent the deterioration in activity of the protease.
