PREPARATION OF TISSUE CULTURE PRODUCT OF BRAZILIAN CARROT

PURPOSE:To produce large amount of valuable substance such as crude saponin, crude saponigen, beta-ecdysone, etc., by the tissue culture of callus derived from living tissue of Brazilian carrot or a root which is a redifferentiated organ of the callus using a specific medium. CONSTITUTION:Callus obtained from living tissue of Brazilian carrot such as Pfaffia paniculata, Pfaffia iresinoides, etc., or a root obtained as a redifferentiated organ of the callus is cultured at 15-40 deg.C in dark place for 14-240 days in a medium produced by adding 0.1-10mg of 2,4- dichlorophenoxyacetic acid, 0.1-10mg of indoleacetic acid, 0.01-5mg of kinetin and/or 0.01-5mg of indolebutyric acid to a basal medium containing inorganic salt, vitamins, saccharose, agar, coconut milk, etc. Crude saponin, crude sapogenin, b-ecdysone, etc., can be produced by this process. The production of callus can be increased and the content of the valuable components can be remarkably increased by this process.
