PHAGE VECTOR K9 FOR CONSTRUCTING OF GENOME LIBRARIES

A phage vector lambda SK9 for construction of genome libraries along identification sections of restrictase BamI and Eco RI has the size of 47.7 k.b.p.; the vector capacity is from 4.5 to 19.0 k.b.p.; the vector consists of lambda EMBL 3 connected to phasmid SK2 DELTA SR which is constituted by phasmid SK2A, from which SalI and Eco RI sections are removed; said phasmid SK2A is made of vector SK18 obtained from DNA fragments of phage lambda , phage M13 and cosmid pHC79; the host spectrum for lambda SK9: LE392 = F',hsd R514(rk,mk),sup E44,sup F58,lac YIOZ(lac IZY)6,gal K2,gal T22,met BI,trp R55, j.M.109 = rec DELTA I, DELTA lac pro,end AI,gyr A96,thi-I,hsd R17,sup E44,rel AI,F,tra D36,pro AB+,lac IqZ DELTA M15. Q359 = hsd Rk-,hsd Mk+,sup F,o 80,P2. The claimed vector was deposited on 21.07.89 at the All-Union Collection of Industrial Microorganisms of the USSR Research Institute for Genetics and Industrial Microorganism Breeding, and was registered under number PH N DEG 687.
