NEW OCTAPEPTIDE AND ANGIOTENSINASE INHIBITOR THEREFROM

NEW MATERIAL:An octapeptide having a peptide structure consisting of L- amino acid sequence of the formula. USE:A therapeutic/preventive medicine for hypertension, therapeutic medicine for heart failure, angiotensinase inhibitor. PREPARATION:For example, deionized water is added to sardine muscle followed by homogenization, and the pH value of the resulting system is adjusted to 2.0 with 1N hydrochloric acid followed by addition of pepsin and then agitation at 37 deg.C for 20hr to effect hydrolysis. The resultant reaction liquor is then subjected to a ultrafiltration membrane and the filtrate is treated in a strongly acidic cation exchange resin column followed by elution with 2N ammonium hydroxide, and the resulting elute is put to vacuum concentration to remove the ammonia contained. The concentrate thus obtained is put to gel filtration, and a fraction having angiotensinase activity is collected and purified by high-performance liquid chromatography, thus obtaining the objective new octapeptide having the peptide structure of the formula.
