PURPOSE:To enable the separation of subprotoplast from pollen of the genus Eucalyptus which has been difficult to be separated from the pollen tube of a pollen having three germ pores. CONSTITUTION:A subprotoplast can be separated from a pollen tube by treating a germinated pollen of the genus Eucalyptus with an enzymatic liquid (cellulase, pectinase, etc., or their combination) adjusted to an osmotic pressure of 0.8-1.2M with an osmotic pressure regulator consisting of 0.5-0.8M of mannitol and 0.2-0.7M of sucrose.
