PURPOSE:To facilitate imbedding of the aimed gene into a chromosome, by forming a variant transformed plasmid having imparted property of integration into the chromosome of a host cell by holding in the host cell and giving a temperature change. CONSTITUTION:A variant plasmid having property of integration into a chromosome of a host cell by holding in the host cell and giving a temperature change is formed. The aimed gene is then recombined in the variant plasmid and a host holding the recombinant plasmid is cultivated. In carrying out the cultivation, the cultivation temperature can be adjusted to readily imbed the aimed gene in the chromosome and stably express the gene. Bacillius stearothermophilus is cited as the host cell and a plasmid pTRA117 shown in the figure may be cited as a specific example of the variant plasmid.
