PURPOSE:To readily remove a free and contaminating coloring matter ligand by purifying an antigenic protein according to triazine coloring matter affinity chromatography and then subjecting the resultant purified antigenic protein to hydrophobic chromatography. CONSTITUTION:A gel of a chromatographic material prepared by covalently bonding a triazine coloring matter to a carrier having a high physical strength is added to an antigenic solution containing an antigenic protein (e.g. pertussis toxin or fibrous hemagglutinin) having specific affinity for the triazine coloring matter and nearly the total amount of the antigenic protein is adsorbed on the aforementioned chromatographic material, which is then washed to remove nonspecifically adsorbed endotoxins. The adsorbed fractions are subsequently eluted to provide an antigenic solution in a high concentration. The above-mentioned antigenic solution is then added to a hydrophobic column to remove the contaminating coloring matter ligand. Thereby, the coloring matter ligand can be completely removed without using any expensive apparatus, such as an ultracentrifuge, and deterioration in yield due to insolubilization of the antigenic protein can be avoided.
