RECOMBINANT PLASMID DNA PVH 64 INTENDED FOR MAPPING OF SMALLPOX VIRUS GENES OF LIVP-STRAIN AND FOR DIRECTED INSERTION OF FOREIGN INFORMATION IN SMALLPOX VIRUS GENOME, AND A METHOD OF ITS CONSTRUCTION
RECOMBINANT PLASMID DNA PVH 64 INTENDED FOR MAPPING OF SMALLPOX VIRUS GENES OF LIVP-STRAIN AND FOR DIRECTED INSERTION OF FOREIGN INFORMATION IN SMALLPOX VIRUS GENOME, AND A METHOD OF ITS CONSTRUCTION
FIELD: biotechnology, genetic engineering. SUBSTANCE: recombinant plasmid pVH 64 (size is 5804 nucleotide pairs) contains vector part of plasmide pBR 322 and Hind III-P-fragment of DNA of smallpox virus, strain LIVP. Cloning Hind III-P-fragment contains genes of proteins 36k and 12k of smallpox virus, strain LIVP. Vector plasmide contains bla-gene providing resistance to ampicillin. Prepared plasmide is used for mapping of viral genes and as a molecular probe for detection of homologous fragments of any orthopox-viruses. Recombinant plasmide pVH 64 can be used for preparing of bacterial producers of viral proteins, and as a vector for incorporation of foreign information into viral genome. Nucleotide sequence of Hind III-P-fragment is determined. EFFECT: preparing of recombinant plasmid DNA indicated above. 3 cl, 1 tbl
