FREEZING AND PRESERVATION OF PRIMORDIAL GERM CELL AND GERM CELL

PURPOSE: To provide a method for freezing and preserving a primordial germ cell or a germ cell of a bird to use it as a material for developmental engineering or preservation of species. ;CONSTITUTION: A primordial germ cell separated from an embryo of a bird or a germ cell before meiosis is floated in a freezing medium containing a freezing and protecting substance (e.g. DMSO, glycerol or ethylene glycol) and put in a tube for freezing. The temperature of the tube is dropped at 1°C/minute rate of cooling up to -80°C liquid nitrogen at -196°C is added and the tube is preserved in liquid nitrogen. A primordial germ cell separated from a genital lunate ring of a chicken embryo at stage 4-12 or separated from blood of an early embryo of a chicken at stage 12-18 is preferable as the primordial germ cell. A primordial germ cell or a germ cell separated from a gonad germ or a gonad of a chicken embryo at stage 23-26 are preferable as the primordial germ cell or the germ cell. This method is expected to have effects such as relief of a scarce species in birds, preparation of transgenic animal, preparation of a new genetic marker animal at a cell level and preparation of disease model animal.;COPYRIGHT: (C)1996,JPO
