DETERMINATION OF CANCER OF COLON AND CANCER OF RECTUM BY MEASUREMENT OF BILE ACIDS IN SERUM BY ELISA

PROBLEM TO BE SOLVED: To provide a method for determining cancer diseases of the colon and the rectum, by measuring bile acids in serum by ELISA (enzyme linked immunosolvent assay). ;SOLUTION: Active ester of bile acid is produced to be reacted with a bovine serum albumin soln. to be dialyzed, and a mammal other than a human being is immunized with the dialyzed substance used as an antigen to prepare anti-bile acid antibody. The above mentioned active ester is reacted with enzyme to form enzyme labelled bile acid as an enzyme labelled antigen. A diluted soln. of serum to be measured, an anti-bile acid antibody soln. and an enzyme labelled antigen soln. are added to a second antibody solid phase plate to be reacted, and a substrate is further added to be reacted. The absorbancy of this reaction product is measured and the concn. of each of bile acids is calculated from a simultaneously measured standard curve. From the concns. of obtained deoxycholic acid(DCA) and cholic acid(CA), the concn. ratio (DCA/CA) of them is calculated and this obtained DCA/CA ratio is compared with the reference range of this ratio of a healthy person to determine cancer of the colon and cancer of the rectum.;COPYRIGHT: (C)1999,JPO
