The invention concerns a method for preparing true acetylenic compounds by reacting lithium monoacetylide with an electrophilic reagent, characterised in that it is brought to a reaction temperature between -30 °C and +30 °C and it consists in: saturating a tetrahydrofurane (THF) solution in acetylene (HC CH); gradually adding a n-butyllithium (n-BuLi) solution to the acetylene-saturated tetrahydrofurane solution; re-saturating in acetylene the mixture resulting from the previous step, at the end of the addition of n-butyllithium; gradually adding said electrophilic reagent (RX) to the mixture re-saturated in n-butyllithium so as to obtain a reaction mixture leading to the formation of said true acetylenic compound.
