PROBLEM TO BE SOLVED: To provide a method for efficiently isolating/purifying in a short time with a simple column operation chib 2-type chitinase as a new class belonging to PR-8 chitinase family from soybeans as raw material. ;SOLUTION: This method comprises the following steps: defatted soybean flour is subjected to extraction with deionized water or an acetate buffer solution, a protein fraction is separated from the resultant extract solution and then put to dialysis treatment followed by hydrophobic chromatography, a fraction having chitinase activity is then separated from the resultant eluate and subsequently subjected to hydrophobic chromatography again. The other version of this method comprises the following practice: the purified chib 2-type chitinase obtained above is passed through a column with mannose-bindable lectin bound to an insoluble support to effect separation into sugar chain-bearing and sugar chain-free modifications of the further purified chib 2-type chitinase.;COPYRIGHT: (C)2001,JPO
