Mixing technique of Umbilical Cord Bloods without immunreaction in case of mixing between mixed Umbilical Cord Blood and Peripheral Blood

PURPOSE: A method for mixing the mixed cord blood(CB) with the peripheral blood without causing immune reaction is provided, which improves the availability of the cord blood containing a small amount of blood stem cells that are essential in treatment of aplastic anemia and leukemia. CONSTITUTION: The method comprises the steps of: determining the HLA-ABDR antigen types of cord blood(CB) and peripheral blood by serological method and DNA serum method; selecting the testing examples, of which all of 6 antigens are different each other, from the testing groups having the same HLA antigens; performing mixed lymphocyte culture with the selected examples by incubating them in 5% CO2 incubator at 37 deg.C for 7 days with irradiating radioactive ray into CB or peripheral blood; measuring the degree of immune reaction of the testing samples; and determining samples causing no immune reaction, in which at least one of HLA-DR antigens correspond between the mixed core blood and peripheral blood, if the samples causes no immune reaction.
