A procedure for the modification of the expression of a Nucleic Acid Sequence present endogenously in a eukaryotic cell.Procedure comprises the steps: a) the cell is transfected with a Vector comprising (i) at least ecuencia Nucleic Acid that binds a Protein Activator.(ii) a positive Selection Marker Gene, (iii) DNA sequence homologous to a segment of nucleic acid in the cell Genome, DNA flanking lassecuencias (i) and (ii)Parapermit Go a homologous recombination, b) the cell transfected is cultivated under conditions under which it is produced unarecombinacion homologous vectorAnd (c) the cell obtained by Stage (b) is recovered. The Invention also refers to a vecto R to be used with the procedure and a eukaryotic Cell obtained by the same.
