The present invention provides non-radioactively labeled synthetic substratesfor enzymatic reactions which exhibit markedly improved solubility having thegeneral structure *F-R1-L1-R2-PHc1-PS-PHc2-(R3-L-R4-T)y. These substrates maybe designed to carry a charge to allow electrophoretic separation ofsubstrates and reaction products. The invention also provides enzymaticactivity assays for protein kinases, phosphatases and proteases utilizing thesubstrates of the invention, as well as methods of producing these substrates.In addition, the invention also provides libraries of the substrates, andmethods of utilizing these libraries to select optimal synthetic peptideenzyme substrates for high-throughput screening assays.
