FIELD: biotechnology, biochemistry, enzymes. SUBSTANCE: method of isolation of superoxide dismutase from yeast Saccharomyces cerevisiae involves disruption of cells of producer and isolation of an active component by treatment of cells of producer. Multistage chromatography is carried out successively on sorbents Soloza, Sp-Sephadex and C8--cellulose. Chromatography on Soloza KG is carried out in static regime at pH 4.0-4.2 in 0.1 M solution of sodium dihydrogen phosphate and elution of the end product at pH 5.0-5.2. Chromatography on Sp-Sephadex is carried out on sorbent equilibrated with 0.1 M solution of sodium dihydrogen phosphate and elution of superoxide dismutase is performed with 0.1 M an aqueous solution of sodium hydrogen phosphate. Chromatography on C8-cellulose is carried out at pH 3.9-4.1 using phosphate buffer solution as an eluent. The proposed technology ensures to obtain the preparation of superoxide dismutase with specific activity about 500 000 U/mg and high yield. Invention can be used in medicine, veterinary science and allied sciences and technique for superoxide dismutase preparing.
