Preparation of an antibody (Ab1; immunoglobulin Y DELTA Fc) from egg yolk that is practically free from an Fc-equivalent region, is new. Preparation of an antibody (Ab1; immunoglobulin Y DELTA Fc) from egg yolk that is practically free from an Fc-equivalent region comprising: (1) immunizing a female poultry bird with an antigen (Ag) so that Ag-specific antibodies accumulate in the yolk; (2) recovering the antibodies; (3) removing non-aqueous biomolecules and granules to produce an antibody-containing water soluble fraction; (4) passing through an inert carrier matrix of immobilized Ag at pH 4-7 and ionic strength below 50 mM to form an immune complex; (5) dissociating the immune complex and eluting Ab1, is new. Independent claims are also included for the following: (1) Ab1 produced this way; (2) immunoassay kit or reagent that contains Ab1; (3) single radial immunodiffusion assay that uses an egg yolk antibody; (4) immunoassay for qualitative or quantitative analysis of an etiological agent that involves treating a sample, at pH 4-7 and ion strength below 50 mM, with an antibody from egg yolk; and (5) immunoassay kit containing an Ag-specific egg yolk antibody, where the assay is performed at pH 4-7 and ionic strength below 50 mM.
