A sample is extracted, desorbed and injected into a high performance liquid chromatography apparatus. This is carried out automatically, with on-line coupling. An Independent claim is included for the processing vessel used, comprising an outer vessel (1) with a glass insert (2), and a closure (6) with septum (8). The glass insert (2) has a hollow cylindrical lower section (4). Its upper section (3) widens upwardly towards the neck (11), which it enters axially. Preferred Features: Analytes are extracted from the sample using fibers. The analytes are then desorbed in a volume less than 30 ml. The extract is injected into the high performance liquid chromatography apparatus. In the apparatus construction, the lower part of the glass insert carries a resilient spacer (5) in contact with the vessel base (9) and/or vessel wall. The volume of the cylindrical lower section of the insert is less than 30 mu l.
