PURPOSE: Provided is a method for efficiently mass-producing beta-chitosan having excellent physical property by deashing and deproteinizing a cartilage of cuttlefish, and deacetylating the cartilage under vacuum condition. CONSTITUTION: The method comprises the steps of (i) reacting a cartilage of cuttlefish with aqueous, inorganic strong acid at 40-95 deg.C to remove ash from the cartilage; (ii) reacting the cartilage with aqueous base at 75-95 deg.C twice or more, so as to remove protein from the cartilage; and (iii) reacting the cartilage with aqueous base at 85-95 deg.C twice or more to deacetylate the cartilage. The deashing process of step(i) is effected by reacting the cartilage with 0.5-1 N HCl for 2-5 hours. The deproteinizing process is performed by reacting the cartilage with 0.5-1 N NaOH for 3-5 hours.
