Chicken Newcastle Disease Virus Nucleocapsid Protein and Method for producing thereof in E.coli

A recombinant nucleocapsid protein of chicken newcastle disease virus and a method for producing the same protein in Escherichia coli are provided, which recombinant nucleocapsid protein has improved reactivity to an antibody against the chicken newcastle disease virus in the serum, so that it can be useful for detecting presence and titer of the virus antibody in poultry, and is easily produced and purified by using E. coli that is simply cultured. The recombinant nucleocapsid protein of chicken newcastle disease virus has the amino acid sequence set forth in SEQ ID NO:1. A gene encoding the recombinant nucleocapsid protein of chicken newcastle disease virus has the nucleotide sequence set forth in SEQ ID NO:4. A recombinant expression vector pET28 comprises the gene encoding the recombinant nucleocapsid protein of chicken newcastle disease virus of SEQ ID NO:4 which is operably linked to the vector. A transformed E. coli BL21 is produced with the recombinant expression vector pET28. The method for producing the recombinant nucleocapsid protein of chicken newcastle disease virus comprises the steps of: (1) preparing the gene encoding the recombinant nucleocapsid protein of chicken newcastle disease virus; (2) constructing a recombinant expression vector comprising the gene encoding the recombinant nucleocapsid protein of chicken newcastle disease virus which is operably linked to the vector; (3) transforming E. coli with the recombinant expression vector; and (4) culturing the transformed E. coli and isolating and purifying the recombinant nucleocapsid protein of chicken newcastle disease virus from the cultured medium.
