A composition comprising 1,2-dioctanoyl-sn-glycerol 3-phosphate is provided to be able to effectively differentiate acute myelocytic leukemia(APL) cell into dendritic cell having strong antigen presenting capacity, thereby being able to be used for autologous cell therapy. The composition for inducing differentiation of APL cell into dendritic cell comprises 1,2-dioctanoyl-sn-glycerol 3-phosphate as an effective ingredient, specifically in the concentration of 100-300 micromoles. The method comprises the steps of: (a) treating the APL cell with the 1,2-dioctanoyl-sn-glycerol 3-phosphate so as to differentiate it into unripened dendritic cell; and (b) treating the unripened dendritic cell with TNF-alpha, lipopolysaccharide(LPS) or a mixture thereof.
