A red pepper CaPO2 gene, and a screening method of resistance against plant disease and environmental stress and transgenic plants by using the same gene are provided to promote development of a new plant variety having resistance against plant disease and environmental stress. The red pepper CaPO2 gene is isolated from Capsicum annuum L. cv. Hanbyul and has the nucleotide sequence of SEQ ID NO:1. A plant disease-resistant protein encoded by the CAPO2 gene has the amino acid sequence of SEQ ID NO:2. The transgenic Arabidopsis thaliana is produced by transforming Arabidopsis thaliana with a recombinant vector pBIN35S: :CaPO2 which is produced by inserting the CaPO2 gene into a vector pBIN35S. The screening method of resistance against plant disease and environmental stress comprises the steps of: isolating RNA from a red pepper plant inoculated by a pathogenic or nonpathogenic bacterium of Xanthomonas campestris pv. vesicatoria; subjecting the isolated RNA to electrophoresis; transferring the developed RNA to a nylon membrane(Hybond N+); and reacting the RNA with a CaPO2 gene probe treated with 32P.
