AUREUS; EXPRESSION VECTOR; RECOMBINANT HOST CELL; METHOD FOR PRODUCING AN ALTERED FNB; METHOD FOR INDUCING AN IMMUNOLOGICAL RESPONSE IN A VERTEBRATE; ISOLATED, CHANGED STAPHYLQCOCCAL AUREUS 3. AUREUS FIBRONECTIN (FNB) BINDING PROTEIN; AND NUCLEIC ACID ISOLATED MOLECULE ENCODING A CHANGE OF 3. AUREUS FNB An isolated, altered 3. aureus protein (Fnb) is described that has at least one mutation in an amino acid selected from residues corresponding to Gln103, Gln105, Lys157, Lys503, Lys620, Lys702, Lys762, Gln783 and Gln830 of the S. aureus strain ATCC49S525. Substitution of these reactive residues within the fibronectin binding protein makes this protein less capable than wild-type covalently cross-linked Fnb with fibronectin and fibrin. Altered fibronectin binding protein effectively interferes with S. aureus adhesion to fibronectin and fibrin and, consequently, an immunogenic composition comprising such altered Fnb exhibits improved immunogenic properties and is safer to use.
