A method for concentrating and desalting a peptide sample is provided to simply concentrate, wash and isolate the peptide sample at low concentration existing in a large amount of a solution without damage, thereby being used as a pre-treating method for a highly sensitive mass-analysis of the peptide sample. A method for concentrating and desalting a peptide sample comprises the steps of: (a) absorbing the peptide sample having a molecular weight of 500-5,000 into a nano-porous carbon foam to concentrate it; (b) washing the sample-absorbed nano-porous carbon form with trifluoroacetic acid, formic acid or acetic acid to remove salts therefrom; and (c) isolating the desalted concentrated sample using an elution solvent such as at least one organic solvent selected from the group consisting of acetonitrile(ACN), acetone, MeOH, EtOH, isopropanol, dichloromethane, ethyl ether, hexane and dimethylformamide, at least one acid selected from the group consisting of trifluoroacetic acid, formic acid, acetic acid, phosphoric acid, citric acid and heptafluorobutyric acid, and a mixture thereof.
