PPROBLEM TO BE SOLVED: To solve the problem wherein a multidimensional liquid chromatography is effective in the separation of a complicated sample like a bio-sample, but only proteome such as protein is applied in a two-dimensional liquid chromatography (2D-HPLC) to which an existing system [ion exchange (IEX)-reverse phase (RP)] column is connected, while, in the separation analysis of a large-hydrophylicity component (e.g., sugar chain or sugar peptide), the adaptation of the reverse phase two-dimensional liquid chromatography is difficult. PSOLUTION: An HILLIC mode is allowed to function by an acetonitrile solvent (50% or above) using an IEX column and an HILLIC column to separate and analyze the large-hydrophylicity component (e.g., sugar chain or sugar peptide) by an on-line 2D (IEX-HILLIC)HPLC system to which the IEX-HILLIC column is connected.
