This invention relates to a liposome consisting of a chloroform soluble and extractable single polar lipid from a mycobacterium wherein said chloroform soluble and extractable single polar lipid is selected from the group consisting of phosphatidylinositol (PI), phosphatidylinositol mannoside (PIM1), phosphatidylinositol dimannoside (PIM2), a mono or dipalmitoylated forms of PIM1 or PIM2, and an acylated-phospholipid having a fast atom bombardment mass spectrometry (FAB MS) spectrum peak at 899, 1139, or 1155 m/z, wherein the liposome, when in the form of an adjuvant composition with an antigen, has immunostimulatory activity. The invention also relates to a method of preparing the liposome.
