PROBLEM TO BE SOLVED: To provide a separation method of a hydrophilic organic compound which can achieve excellent separation, even when a highly hydrophilic high-polarity compound is used as a separation object, and to provide a filler for hydrophilic interaction chromatography.SOLUTION: In the separation method of the hydrophilic organic compound, the hydrophilic organic compound is separated from a raw material by a hydrophilic interaction chromatography method, by using as a stationary phase, surface-modified silica gel wherein the surface of silica gel is modified by a functional group shown by general formula (I) (wherein R, R, Rare each hydrocarbon group having the carbon number of 1-11, or each hydrocarbon group in which a part of the hydrocarbon group is substituted by one or two or more kinds of atomic groups selected from alkoxy, carbonyl, amino, amide and carboxy, or an atomic group is inserted into a carbon chain).
