A method for improving the yield of vitamin C precursor 2-keto-L-gulonic acid by means of mixing strains to create a subculture and by enhancing the interaction between two strains, said method including steps for solid culturing, seed culturing, isolation and purification, and fermentation. Also provided is a method for using the mixed subculture to detect changes in protein, phospholipid component, nutritional environment and micromolecular metabolite in the process of passage of strains for industrial production of vitamin C. In addition, the present invention also comprises a use for gluconobacter oxydans having a vector encoding a sorbosone dehydrogenase gene; a method for detecting change in protein in cells in the process of applying glutathione to gluconobacter oxydans; and a method for a quality control index for the use of corn steep liquor as a raw material for fermentation.
