Provided is a preparation method for inductive pluripotent stem cells, comprising the following steps: step 1, introducing one or multiple stem cell pluripotent factors into somatic cells; step 2, culturing said somatic cells prepared in step 1, in the process, further using a hyperosmotic stress culture medium to culture said cells; step 3, identifying inductive pluripotent stem cell clones. Also provided is a culture medium for preparing inductive pluripotent stem cells, the culture medium having hyperosmotic stress being used in the preparation of iPS cells. The hyperosmotic stress environment can increase the production efficiency of mouse iPS cells 10 to 25 times. The iPS cell induction method is conducive to the enhancement of the safety of iPS technology and the use of iPS cells in the field of regenerative medicine.
