PROBLEM TO BE SOLVED: To provide a method by which a large-scale long-chain DNA-type RNAi library can be prepared by once to several-time operations under the same preparation condition.;SOLUTION: The recombinant vector including a reverse reiterated sequence originated from a foreign DNA fragment is prepared by utilizing a nucleic acid in which one strand of a double strand contains a nicking site of the first nicking endonuclease, an insertion region of the foreign DNA fragment, a nicking site of the second nicking endonuclease, the reverse reiterated sequence sandwitching a spacer region, and a nicking site of the first restriction enzyme in the direction of from 5' to 3'.;COPYRIGHT: (C)2010,JPO&INPIT
