The present invention relates to an insulin-secreting peptide derivative having a modified N-terminal charge and a pharmaceutical composition comprising the same. Specifically, the insulin-secreting peptide derivative has a positive charge at the N-terminal of the insulin-secreting peptide. It is characterized by being modified to neutral charge or net negative charge at neutral pH. The insulin-secreting peptide derivative according to the present invention is a natural insulin-secreting peptide that is rapidly dissociated from the GLP-1 receptor by maintaining the N-terminal charge as described above, maintaining blood stability. Since it exhibits increased insulin secretion ability and blood glucose level lowering ability, it can be used very effectively for the treatment of type 2 diabetes.
