Large scale generation of functional megakaryocytes and platelets from human embryonic stem cells under stroma free conditions. The present invention provides a method for generating megakaryocytes and platelets. In several embodiments, the method involves the use of hemangioblast-derived human embryonic stem cells for differentiation into megakaryocytes and platelets under serum and stroma free condition. In this system, ctehs are conducted toward megakaryocytes through embryoid body formation and hemangioblast differentiation. Additionally, a method for treating a subject requiring platelet transfusion is provided.
