The present invention relates to an anti-inflammatory composition for treating or preventing chronic inflammatory diseases caused by excessive activities of NLRP3 inflammasome, by inhibiting the activities of the NLRP3 inflammasome of macrophages, comprising IL-4 or an IL-4 activator, and relates to a screening method thereof. According to the present invention, the anti-inflammatory composition has excellent effects in treating or alleviating inflammation, thereby being broadly used as a therapeutic agent for inflammation or a supplementary food for health comprising the same as an active ingredient. According to the present invention, the screening method can be efficiently used for developing the therapeutic agent for inflammation and for studying the mechanism of IL-4 action.
