The present invention relates to a method for isolating extracellular endoplasmic reticulum using an aqueous two phase system. The method for isolating extracellular endoplasmic reticulum using an aqueous two phase system according to the present invention comprises: (a) a step of mixing a first material and a second material which are not blended with each other with an aqueous solution containing body fluid or extracellular endoplasmic reticulum to prepare the aqueous two phase system; and (b) a step of isolating the extracellular endoplasmic reticulum concentrated in the second material from the aqueous two phase system. The method for isolating extracellular endoplasmic reticulum using an aqueous two phase system according to the present invention has merits that a very high efficiency is obtained, an isolation method is simple, and the time required is very short compared to the prior art. More specifically, isolation of the extracellular endoplasmic reticulum using the aqueous two phase system does not require an ultracentrifuge, and the isolation of the extracellular endoplasmic reticulum is performed almost to an efficiency of 100% within a short time of about 10 to 20 minutes. Therefore, a method for isolating an extracellular endoplasmic reticulum using an aqueous two phase system according to the present invention is a method which is expected to have a positive influence on diagnostic development using the extracellular endoplasmic reticulum, and of which competitiveness is very high since the method for isolating the extracellular endoplasmic reticulum using the aqueous two phase system according to the present invention is practical and incurs low expenses such that the method is economical differently from existing other isolation methods. Further, there are merits that a method for isolating an extracellular endoplasmic reticulum using an aqueous two phase system according to the present invention is capable of increasing purity of an extracellular endoplasmic reticulum contaminated by protein, is capable of diagnosing diseases by the isolated extracellular endoplasmic reticulum, and is capable of being applied to all fields using the extracellular endoplasmic reticulum.
