The present invention provides a bioprocess for production and purification of enzyme α- L-rhamnosidase from yeast strain Clavispora lusitaniae for the conversion of bitter naringin to nonbitter rhamnose and prunin in citrus juice. The α-L-rhamnosidase is used to convert bitter naringin to nonbitter prunin and rhamnose. Yeast strain Clavispora lusitaniae is capable of producing α-L- rhamnosidase enzyme by hydrolyzing naringin and showed maximum activity in presence of rhamnose (0.6% w/v), yeast extract (0.4% w/v), temperature (35 °C) and pH (4) in the minimal medium supplemented with naringin (0.2% w/v).
