By a high-throughput sequencing means, high-throughput sequencing is respectively performed on mixed culture ponds of female individuals and male individuals, a sequencing result is bioinformatically analysed to screen out significantly differential sequence fragments in the female-male mixed culture ponds, the obtained differential fragments are verified in individuals of different sources, a probe sequence which can be used to distinguish female Litopenaeus vannamei from male Litopenaeus vannamei is obtained, and the sequence can be utilized to carry out accurate determination of the genetic sexes of this type of shrimp.
