Compositions and methods for the administration of progestin hormones having affinity for binding to sex hormone binding globulin (shbg) are disclosed. The compositions combine such progestins with non-progestin shbg ligands to displace at least part of the shbg progestin in blood plasma, thereby increasing their bioavailability. Methods of modulating blood progestin and estrogen levels through the use of shbg binding and displacement to optimize the effectiveness of contraceptive formulations and minimize side effects and adverse events are also disclosed.
