A method for extracting polyphenol substances from semen juglandis with inner seed coats by the aid of combined enzyme and ultrasonic treatment. The method comprises: obtaining semen juglandis with seed coats; feeding the semen juglandis with inner seed coats into an ethanol aqueous solution according to a volume ratio of 1:3-1:15, regulating the pH of the mixture by the aid of food-grade citric acid or hydrochloric acid until the pH of the mixture is 3.5-6.5, and then adding pectinase and/or a cellulase compound to obtain a polyphenol extracted system. According to the semen juglandis polyphenol crude extract prepared by the method, the total weight of dry substances, which are obtained after freeze drying, at least accounts for 2% of the weight of the original semen juglandis with the coats, and the contents of polyphenol effective substances in the semen juglandis polyphenol crude extract can reach 15% at least; the semen juglandis polyphenol crude extract further can be separated by the aid of macroporous adsorption resin and industrial chromatographs, and accordingly products such as ellagic acid, hydrolysable tannin, and juglone can be prepared.
