The present invention relates to a signal amplification method in a lateral flow analysis using nanofibers having a core-sheath structure, and a lateral flow analysis device using the same. More specifically, the present invention relates to a signal amplification method in a lateral flow analysis and a lateral flow analysis device configured to perform the method of the present invention, wherein the signal amplification method comprises the steps of: binding an analyte and a conjugate having a first antibody or a first specific binding material that can specifically bind to a first epitope or a first binding site of the analyte bound to a label; binding the analyte to which the conjugate is bound, and a second antibody or a second specific binding material, which is immobilized, specifically binding to a second epitope or a second binding site of the analyte; and bringing, into a reaction, a signal amplification material, which is delayed, to be transmitted, in a signal amplifying part formed of double layered nanofibers of a sheath part made of water-soluble materials and a core part made of signal amplifying materials.
