The present invention relates to a three-dimensional cell culture system comprising polyethylene glycol (PEG)-based hydrogel. The PEG-based hydrogel is prepared by mixing polyethylene glycol-vinylsulfone (PEG-VS), which is PEG bound to vinylsulfone, adhesion peptide, and a crosslinker. The system can culture dermal fibroblast, melanocytes, or keratin cells, which are skin cells, and thus can be advantageously used to measure the activity of skin cells.
