The present invention relates to a microfluidic device for a differential pressure viscometer, which is used for measuring extensional viscosity of various fluids including a bio-related fluid such as mucus, joint fluid, or blood, a fluid such as paint or inkjet printer ink, fuel oil, and a polymer solution, and a method for designing the microfluidic device. According to the present invention, the microfluidic device for the differential pressure viscometer includes: a sample microchannel in which an extensional flow occurs at a predetermined extensional velocity; and a comparative microchannel in which a pressure drop identical to a pressure drop due to a shear flow in the sample microchannel occurs, so that the extensional viscosity at a constant extensional velocity (in a steady state) is measured.
