The present invention relates to a polypeptide having improved purity of a protein and affinity for an antigen, a complex with an antibody or an antigen-binding fragment thereof, and manufacturing methods thereof. The polypeptide or the complex thereof of the present invention does not generate glycosylation even when being produced in a eukaryotic cell, so that purity of a produced protein is high. Also, affinity with respect to a target antigen is high, thereby having a very high value as a reagent for diagnosing or treating a disease.
