Provided is a method for cultivating aphid-resistant tobacco and wheat. The method comprises: inhibiting the expression of a specific gene or specific gene fragment in an initial plant to obtain an aphid-resistant transgenic plant. The specific gene fragment is the aphid-specific target gene 22544 isolated from the aphid's intestinal tract, with the nucleotide sequence thereof as shown in sequence 1, and the specific gene is a gene having the nucleotide sequence as shown in sequence 1. Also provided is a system for cultivating the aphid-resistant transgenic plant, comprising an interference fragment or interference vector that inhibits the expression of the specific gene or specific gene fragment in the initial plant. The transgenic plant obtained by introducing the target gene 22544 as the target, wherein the expression of same is to be inhibited, into the interference fragment or interference vector exhibits a good aphid-resistant phenotype.
