Provided are an automated intelligent detection instrument for thin-layer chromatography and a real-time online detection method thereof, the detection instrument comprising a thin-layer chromatography plate box (13), a liquid phase tank box (14) for development and color developing, a rotary driving device (1) for moving a thin-layer chromatography plate (10), an infusion pump developing solvent injection device, an infusion pump sample sampling and application device (2), and a drying and image capturing apparatus. An automatic detection method is provided. The detection instrument can automatically control steps such as sampling, sample application, development, color developing and scanning in thin-layer chromatography, carry out online qualitative or quantitative analysis on samples in real time, and track, in real time, the change of sample components over time and the progress of a chemical reaction, thereby saving on human labor, improving efficiency and reducing human errors.
