The present invention relates to a water soluble overexpression and purification method of recombinant human serum albumin. More specifically, the present invention provides: a recombinant expression vector including a gene of encoding a water soluble enhancement tag and a human serum albumin gene; a host cell transformed with the recombinant expression vector; a method of producing recombinant protein using the same; and recombinant human serum albumin produced by the method. According to the present invention, the method for producing the recombinant expression vector or recombinant human serum albumin (rHSA) is able to improve the water soluble expression efficiency and productivity of recombinant human serum albumin. The recombinant human serum albumin obtained by the method is expected to play an important role as a broad therapeutic agent for blood related diseases.
